Phenyl-stabilisierte Mono- und Bis-Mercapto-Polyene als trifunktionelle Raman-Reportermoleküle für die oberflächenverstärkte Raman-Spektroskopie

In der vorliegenden Arbeit werden maßgeschneiderte, trifunktionelle und Phenyl-stabilisierte Mono- und Bis-Mercapto-Polyene als Raman-Reporter-Moleküle für die oberflächenverstärkte Raman-Spektroskopie (SERS) vorgestellt. Diese Moleküle weisen ein leicht polarisierbares, ausgedehntes π-System auf und verfügen über eine terminale Thiol- bzw. Esterfunktion. An dieser Stelle wird eine modulare Synthese von Monothiol-Polyenen (MtP) mit Kettenlängen bis zu 7 C=C-Doppelbindungen gezeigt. Die hergestellten MtP werden theoriegeleitet mittels UV/Vis-Extinktions-, Kernspinresonanz-, Raman- sowie SERS-Spektroskopie charakterisiert und auf eine Multiplexing-Eignung überprüft. Beispielhaft wurde mit zwei spektral eindeutig unterscheidbaren Raman-Reporter-Molekülen ein 2-Farben immunchromatographischer Test für eine Paralleldetektion durchgeführt. Ein solcher Test gehört zu den am häufigsten in der patientennahen Labordiagnostik (POCT) durchgeführten Tests, da diese schnell, kostengünstig und zuverlässig durchzuführen sind. Die hierfür verwendeten Polyene unterscheiden sich allerdings in ihrem Adsorptionsverhalten und ihrer spektralen Helligkeit. Eine Verbesserung und Erweiterung der Polyen-Multiplexing-Kapazität kann durch Verwendung von Isotopologe erreicht werden. Im weiteren Verlauf der Arbeit wird die Synthese der Polyen-Isotopologen mit 1 und 2 C=C-Doppelbindungen vorgestellt. Für eine Evaluierung der Multiplexing-Eigenschaften wurden ebenfalls theoriegeleitet Raman- und SERS-Spektren aufgenommen. Ein weiteres Projekt war die Erstellung von trifunktionellen Raman-aktiven Moleküle für eine elektrostatische Assemblierung von Goldnanopartikeln zu Gold/Gold-Kern/Satelliten- Superstrukturen. Bisher wurden diese Superstrukturen mit einem ionischen nicht-Raman-aktiven Alkanthiol hergestellt. Für eine Raman-Signalgebung muss hier ein entsprechender Raman-Reporter co-adsorbiert werden. Mit einem elektrostatisch verbrückenden Raman-aktiven Molekül kann das detektierbare Raman-Signal deutlich verstärkt werden, da in diesem Fall jedes vorhandene Molekül signalgebend ist. Die Weiterentwicklung dieses Konzepts sind Bisthiol-Polyene, die Goldpartikelüber eine kovalente Bindung der Thiolfunktionen molekular miteinander verbrücken. Bisher wurden für die Kopplung von sphärischen Goldnanopartikeln bisthiolische Alkylketten verwendet, mit denen der Abstand zwischen den Partikeln über die Länge des Alkans vorgegeben werden kann. Wie auch bei den Superstrukturen muss ein Raman-aktives Molekül verwendet werden. Da die Synthese der Polyene modular ist, kann der Terminus der zu erfüllenden Aufgabe entsprechend angepasst werden und beispielsweise eine weitere Thiolfunktion endständig eingeführt werden, wodurch ein Raman-aktives Brückenmolekül erhalten wird. In dieser Arbeit wird die Synthese von Bisthiol-Polyenen (BtP) mit 2 und 3 C=C-Doppelbindungen vorgestellt, welche anschließend zur erfolgreichen Assemblierung von sogenannten idealen Dimeren aus jeweils 50 nm großen super-sphärischen Goldnanopartikeln erfolgte und mittels SERS-Spektroskopie charakterisiert wurden.

Tailormade, trifunctional and phenyl-stabilized mono- and bis-thiolated polyenes are presented as Raman reporter molecules for application in surface-enhanced Raman scattering (SERS). These polyenes possess highly polarizable moieties like conjugated π-systems. In addition they exhibit, in case of the mono-thiolated polyene (MtP), a terminal thiol and an ester moiety respectively, in case of the bis-thiolated (BtP) molecule, two thiol moities. In this thesis a modular synthesis of
MtPs with a chain length up to 7 C=C double bonds (DB) is shown. The MtP are experimentally characterized  by  UV/Vis-extinction,  nuclear  spin  resonance,  Raman  and  SERS  spectroscopy complemented by density functional theory calculations to validate their multiplexing capability. As an example, a 2-color immunochromatographic test for parallel detection was carried out with the
two spectrally distinguishable Raman reporter molecules with 2 and 5 DB. Such a test is one of the most frequently performed tests in point-of-care-testing (POCT), since they are fast, cheap and reliable. The polyenes used for this, however, differ in their adsorption behavior and their spectral brightness. An improvement and expansion of the polyene multiplexing capacity can  be achieved  by  using  isotopologues.  In  the  further  course  of  the  work,  the  synthesis  of
polyene isotopologues with 1 and 2 C=C double bonds is presented. To evaluate the multiplexing properties, theory-guided Raman and SERS spectra were recorded.
Another  project  part  included  the  synthesis  of  trifunctional  Raman-active  molecules  for an electrostatic assembly of gold nanoparticles into gold/gold-core/satellite-superstructures. So far, the  superstructures were  made  with an  ionic,  non-Raman-active  alkanethiol.  A corresponding Raman  reporter  has  to  be  co-adsorbed  to  obtain  a  Raman  signal.  With  an electrostatically bridging  Raman-active  molecule,  the  detectable  Raman  signal  can  be significantly  amplified, since every adsorbed molecule present raises the Raman signal.
The further development of this concept are bis-thiol polyenes, which molecularly bridge gold particles via a covalent bond of the thiol functions. So far, bis-thiolic alkyl chains have been used for the coupling of spherical gold nanoparticles, where the distance between the particles can be specified over the length of the used alkane chain. As with the superstructures, an additional Raman-active molecule has to be used. Since the synthesis of the polyenes is highly modular, the
terminus can be adapted accordingly to the task to be fulfilled and, for example, a further thiol function can be introduced at the opposite side, whereby a Raman-active bridge molecule is obtained. In this thesis, the synthesis of bis-thiol polyenes (BtP) with 2 and 3 C=C double bonds is also presented, which were then utilized for the successful assembly of so-called ideal dimers made from 50 nm super-spherical gold nanoparticles. The BtPs were characterized via SERS
spectroscopy due to the dimer assembly.
