Chiralitätsinduktion mittels azolhaltiger C3-symmetrischer Cyclopeptide

Basierend auf der Struktur des C3-symmetrischen marinen Cyclopeptids Westiellamid wurden trifunktionalisierte Analoga konzipiert, die als zentrale Bausteine für künstliche Rezeptoren, Containermoleküle oder Metallliganden verwendet werden können. Diese neuen Makrocyclen verfügen über Hydroxymethyl-, Chlormethyl- bzw. Aminomethyl-Ankergruppen, die direkt an den Oxazol- bzw. N-Methylimidazoleinheiten des starren Grundgerüsts gebunden sind. Die Synthesen der chiralen Plattformen wurden erarbeitet. Die Makrocyclen 1a-b, 2a-b und 3a-b sind in wenigen präparativen Schritten unter Verwendung geeigneter Schutzgruppentechniken und einer Cyclooligomerisation in Gramm-Maßstab zugänglich. Als Nebenprodukte wurden teilweise auch die entsprechenden Cyclooktapeptide mit C4-Symmetrie isoliert. Ab initio Rechnungen der Cyclohexapeptide ergaben eine schalenförmige Sekundärstruktur, bei der der durch die Azolringe definierte Kegelwinkel von der Art des äußeren Heteroatoms abhängig ist. Durch Darstellung und Untersuchung dreiarmiger, sechszähniger Liganden wurde das Maß der Chiralitätsinduktion, die die Plattformen auf ein koordinativ gebundenes Metallzentrum bewirken können, nachgewiesen. Aus der C3-symmetrischen Aminomethyloxazolplattform 3a wurde der Triscatecholat-Ligand 13 hergestellt. Mittels ESI-MS-Messungen und UV/CD-spektrophotometrischen Titrationsexperimenten konnte eine 1:1-Komplexbildung von 13 mit Al3+-, Ga3+-, In3+-, Fe3+- sowie mit Ge4+- und Ti4+-Ionen nachgewiesen werden. Mit Ausnahme des Ti(IV)-Komplexes wiesen die Änderungen im CD-Spektrum durch Koordination auf eine Fixierung der Catecholatbindungsarme in -Konfiguration um das Metallzentrum hin. Durch quantenmechanische Rechnungen konnte eine extrem hohe diastereoselektive Bildung der oktaedrischen Metallkomplexe bestätigt werden. Die Energien der (SSS,)-Konfigurationen sind um 55 88 kJmol-1 tiefer als die der (SSS,)-Stereoisomere. Die berechneten Strukturen zeigen eine beträchtliche Verzerrung der Konformation des Plattformrückgrats in den Metallkomplexen im Vergleich zur Plattform 3a. Durch eine dreifache Williamson-Ethersynthese wurde ausgehend von der Plattform 1a der Ligand 14 mit drei 2,2´-Bipyridyl-Seitenarmen hergestellt. Durch UV- und CD-spektrophotometrische Titrationsexperimente bzw. Job-Plot-Analysen konnte mit Zn2+- und Ni2+-Ionen eine 1:1 Komplexbildung nachgewiesen werden, während mit Cu2+- und Co2+-Ionen eine Komplexbildung gemischter Stöchiometrie festgestellt werden konnte. Alle Trisbipyridyl-Komplexe 14a-d besitzen eine -Konfiguration am Metallzentrum. Ab initio Rechnungen zeigen bei dem Zn2+-Komplex eine niedrige, aber immerhin signifikante Stereodifferenzierung um 12 kJmol-1. Durch dreifache Amidverknüpfung der Triaminoplattformen 3a-b mit dem dreiwertigen Säurechlorid 84b wurden die Triarylphosphinoxide 89a-b sowie die Triarylphosphine 16a-b hergestellt. Durch NOESY-Messungen und QM-Rechnungen konnte nachgewiesen werden, dass bei allen Substanzen die Arylringe um das Phosphorzentrum in helikaler Anordnung und fixiert in der P-Konformation vorliegen. Die Phosphine 16a-b repräsentieren die ersten bekannten konformationsstabilen Triarylphosphine ohne helikale Epimerisierung und stellen eine neue Klasse chiraler Phosphinliganden dar. Darüber hinaus verhalten sich die Substanzen 89a-b und 16a-b als Containermoleküle, indem sie Chloroform bzw. Dichlormethan in dem inneren Hohlraum einschließen. Die Struktur des Einschlusskomplexes mit CHCl3 wurde durch ab initio Rechnungen bestimmt. Die Stabilität der Einschlussverbindung mit Chloroform ist wesentlich höher als mit Dichlormethan. Die leichte Oxidation der Phosphine 16a-b zu den entsprechenden Phosphinoxiden 89a-b wird durch Einschluss von Chloroform inhibiert.

Based on the structure of the C3-symmetric marine cyclopeptide Westiellamide, novel trifunctionalized pseudopeptide scaffolds were designed, which potentially can be used as central components for artificial receptors, container molecules or metal ligands. These macrocycles bear hydroxymethyl, chloromethyl or aminomethyl groups as anchoring sites directly connected to the oxazole or N-methylimidazole units of their rigid backbone. A convenient synthesis of the chiral scaffolds was elaborated. Macrocycles 1a-b, 2a-b and 3a-b can be obtained on gram scale through few preparative stages involving protective group techniques and a cyclooligomerisation step. As side products the corresponding cyclooctapeptides showing C4-symmetry were isolated. Ab initio calculations of the cyclohexapeptides showed a cone-like secondary structure, where the cone-angle defined by the planes of the azole-rings alters with respect to their outer heteroatoms. Next, synthesis and investigation of three-armed hexadental ligands were performed in order to investigate the extent of chirality induction effected by the platforms on a coordinatively bound metal centre. Triscatecholate ligand 13 was synthesized starting from the C3-symmetric aminomethyloxazole platform 3a. ESI-MS measurements and UV/CD-spectrophotometric titration experiments indicated 1:1 complex formation of 13 with Al3+-, Ga3+-, In3+-, Fe3+- as well as with Ge4+- and Ti4+-ions. Changes of the CD-spectra upon coordination showed with exception of the Ti(IV)-complex fixation of the three catecholate binding arms in -configuration around the metal centre. Quantum chemical modeling proved an extreme high diastereoselectivity for the formation of the observed octahedral metal complexes. The calculated energies for the (SSS,)-configuration are of 55 to 88 kJmol 1 lower relative to the (SSS,)-stereoisomer. The obtained structures for metal complexes 13a-f show a considerable distortion of the backbone-conformation with respect to platform 3a. Ligand 14 was synthesized in a threefold Williamson ether synthesis starting from scaffold 1a. UV/CD-spectrophotometric titrations and Job-Plot analysis showed 1:1 complex formation of 14 with Zn2+- and Ni2+-ions, while a mixed stoichiometry could be observed with Cu2+- and Co2+-ions. In all of the tris(bipyridyl) complexes 14a-f the binding arms adopt a helical orientation of -configuration around the central metal ion. Ab initio calculations revealed for the Zn2+-complex a lower, but still significant stereodifferentiation of 12 kJmol 1. In order to demonstrate the applicability of our platforms for predetermining the chirality at a distant covalently bound phosphorous centre, propeller-like triarylphosphine oxides 89a-b and the corresponding triarylphosphines 16a-b were synthesized by coupling scaffolds 3a-b with the tripodal acid chloride 84b. 2D-NOESY experiments and QM-calculations verified, that the arene rings adopt in all compounds a helically fixed P-conformation. Triarylphosphines 16a-b represent the first known conformationally stable triarlyphosphines without helical inversion and can be treated as a new class of chiral phosphine ligands for conceivable applications as catalysts. In addition, substrates 89a-b and 16a-b act as container molecules by enclosing chloroform and dichloromethane in their cavity respectively. The structure of the inclusion complex with CHCl3 was determined by ab initio calculations. The stability of the inclusion complex with chloroform is much higher than with dichloromethane. The otherwise oxidation sensitive phosphines 16a-b are remarkably stabilized by the inclusion of chloroform.
