Supramolekular-funktionalisierte Polymere – Ein modularer Ansatz

Das Ziel dieser Arbeit war es einen modularen Ansatz zu entwickeln und zu etablieren, welcher es ermöglicht GCP- bzw. GCP-ZW-Derivate mit anderen Bausteinen zu verknüpfen und somit unterschiedliche Anwendungen adressieren zu können. Die Entwicklung dieses modularen Ansatzes konnte in dieser Arbeit durch drei erfolgreich abgeschlossene Projekte bestätigt werden.

Im ersten Projekt dieser Arbeit konnten erfolgreich GCP-Zwitterion-Monomere (GCP1 und GCP2) hergestellt werden. Diese Norbornen-basierten Monomere konnten mittels Ringöffnender Metathese-Polymerisation (ROMP) mit einem weiteren Monomer kombiniert werden. Dieses Ergebnis beweist, dass es möglich ist, ROMP als Grundlage für einen modularen Ansatz für GCP-Derivate zu verwenden.

  • Es konnte erfolgreich ein modularer Ansatz für Polymere mittels ROMP entwickelt werden.

 

Der zwitterionische Charakter der GCP-Motive führt dabei zu einer pH-abhängigen Quervernetzung der Polymerketten, welche mit einer strukturellen Änderung einhergeht. Durch Substitution der Monomere, welche mit dem GCP-Monomer kombiniert wurden, konnten die Eigenschaften der Polymere systematisch optimiert werden. Dies ermöglichte eine Optimierung der Wasserlöslichkeit der Polymere. Zusätzlich konnte die pH-abhängige Schaltung der Polymerstruktur durch Variation der Verhältnisse der Monomere verbessert werden. Durch unterschiedliche Untersuchungen (DLS, AFM, TEM) konnten polymerische Nanopartikel bei neutralem pH-Wert nachgewiesen werden, welche auf den Kollaps der Polymerketten durch die Dimerisierung der Zwitterionen zurückzuführen sind. Diese Nanopartikel konnten durch Erhöhung des pH-Wertes aufgebrochen werden und das Polymer wurde in die offenkettige Form überführt. Zusätzlich konnte ein hydrophober Farbstoff (Nilrot) in den Nanopartikeln eingeschlossen werden und zu einem späteren Zeitpunkt durch Zugabe von Base wieder freigesetzt werden. Diese Ergebnisse zeigen, dass das Polymer eine potenzielles Modell als Wirkstofftransporter (drug delivery system) darstellt.

  • Mit Hilfe des modularen Ansatzes konnten Polymer-Nanopartikel hergestellt werden, welche wasserlöslich und pH-schaltbar sind.

Im zweiten Projekt wurden Polymere untersucht, welche auf dem GCP2-Monomer und Monomeren mit Fluoreszenz-Eigenschaften basieren. Die Synthese dieser neuen Monomere verlief erfolgreich, ebenso wie die anschließenden Homo- bzw. Copolymerisationen. Drei der synthetisierten Monomere basierten auf Cyanostilben-Derivaten und wiesen daher AIE-Effekte auf, welche durch unterschiedliche Untersuchungen bestätigt wurden. Ziel war es im nächsten Schritt durch die Quervernetzung des GCP2 Monomers Polymer-Nanopartikel mit pH-responsiven Fluoreszenzeigenschaften zu generieren. Aufgrund der Bildung dieser Partikel sollte es zu einer Aggregation der Cyanostilben-Derivaten kommen, was eine pH-Abhängige Zu- bzw. Abnahme der Fluoreszenz zur Folge haben sollte. Zur Verbesserung der Löslichkeit wurde auch in diesem Projekt das Monomer M4 eingebracht. Dazu wurde ein Tripolymer hergestellt welches aus den Monomeren GCP2, CS3 und M4 bestand. Dieses Polymer konnte erfolgreich in wässrigen Lösungen gelöst werden. Eine Veränderung der Struktur des Polymers in Lösung konnte ebenfalls durch DLS und AFM nachgewiesen wurden. Zusätzlich konnte eine deutliche Fluoreszenz in Lösung beobachtet werden, welche sich jedoch nicht durch pH-Änderungen beeinflussen ließ. Diese Tatsache basiert mit großer Wahrscheinlichkeit auf dem amphiphilen Charakter des Polymer, welcher z.B. eine Mizellenbildung in Wasser begünstigt weshalb die CS-Anteile dauerhaft aggregiert vorliegen. Aufgrund der pH-abhängigen Strukturen des Polymers und dem nachgewiesenen AIE-Effekt der Monomere, stellt dieses Terpolymer ein erstes Modell-System für weitere Optimierungen dar.

  • Es konnten Monomere und Copolymere mit fluoreszenten Eigenschaften (z.B. AIE-Efefkt) hergestellt werden, jedoch konnten diese nicht durch die pH-Abhängige Dimerisierung des GCP-ZW beeinflusst werden.

Das dritte Projekt dieser Arbeit befasste sich mit der Anwendung von GCP-Polymeren als nicht-virale Vektoren in der Gentransfektion. Es wurden drei neue GCP-Monomere synthetisiert, welche im Anschluss zu Homopolymeren oder in Kombination mit dem Monomer M4 zu Copolymeren umgesetzt wurden. Zwei der Homopolymere zeigten positive Ergebnisse bei Transfektionsexperimenten, es konnten jedoch keine ausreichend hohe Effizienzen beobachtet werden. Durch Einbringen des Monomers M4 konnten sowohl die Löslichkeit in Wasser als auch die Effizienz des GT1 basierten Vektors deutlich gesteigert werden. Neben der Kombination mit weiteren Monomeren ermöglichte der modulare Ansatz zusätzlich eine einfache Veränderung der Monomerverhältnisse. Durch Steigerung des GT1-Anteils konnte die Transfektionseffizienz erneut gesteigert werden, während ein erhöhter M4-Anteil zu einer Verringerung der Transfektion sowie Zunahme der Zelltoxizität führte.

  • Die neu entwickelten GCP-Copolymere eignen sich für den Einsatz als nicht-virale Vektoren in der Gentransfektion und konnten durch den modularen Ansatz systematisch optimiert werden.

Aim of this work was to develop and establish a modular approach, which allows a combination of GCP- and GCP-ZW-derivatives with other components to realise different applications. The development of this modular approach was proven in this word by three successfully completed projects.
In the first project of this work, it was possible to successfully synthesize GCP-Zwitterion-monomers (GCP1 und GCP2). These norbornene-based monomers could be combined with another monomer combined via ring-opening metathesis polymerisation. This result proved that it possible to use ROMP as a basis for a modular approach to polymerize GCP-monomers.

  • It was possible to successfully develop a modular approach for polymers via ROMP.

The zwitterionic nature of the GCP-motifs leads to a pH-depending crosslinking of the polymer chains, which is associated with a structural change of the polymer. By substitution of the monomers, which were combined with the GCP-monomer, it was possible to systematically optimise the properties of the polymers. This also allowed an optimisation of the water solubility of the polymers. Additionally, it was possible to improve the pH-depending switching behaviour by variation of the ratios of the monomers. The presence of polymer nanoparticles at neutral pH was proved by different investigations (DLS, AFM, TEM). These observations are based on the collapse of the polymer chains due to the dimerization of the zwitterion. It was possible to break up these particles and to convert the polymer in an open-chained form by an enhancement of the pH-value. Additionally, a hydrophobic dye (Nilered) could be encapsulated in the nanoparticles and released afterwards at a later time by addition of base. These results show that this polymer represents a potential model for a drug delivery system.

  • It was possible to prepare polymeric nanoparticles by mean of the modular approach, which are water-soluble and pH switchable.

In the second project of these work polymers were investigated, which are based on GCP2-monomer and monomers with fluorescent properties. The synthesis of the new monomers was successful, as well as the followings homo- and copolymerisations. Three of the synthesized monomers were based on cyanostilben-derivatives and showed AIE-properties, which could be proved by different investigations. Aim of this project was to achieve pH-responsive fluorescence properties due to the crosslinking of the GCP2-monomer. Based on the formation of polymer nanoparticles an aggregation of the cyanostilben-derivatives should take place which leads to a pH-depending increase or decrease of the fluorescence intensity. To improve the solubility the monomer M4 was also used in this project. Therefore, a tripolymer was synthesized, which consists of the monomers GCP2, CS3 and M4. This polymer was successfully dissolved in aqueous solutions and a structural change caused by different pH-values could also be proved by DLS and AFM measurements. Additionally, a distinct fluorescence was observed in solution, which could not be influenced by pH changes. One explanation for this behaviour could be the amphiphilic fashion of the polymer, which could promote e.g. the formation of micelles in aqueous solutions. This formation would cause a permanent aggregation of the CS-moieties. However, this terpolymer represents a first model system for the planed application because of the pH-depending structures of the polymer in solution and the proved AIE behaviour of the monomers.

  • Monomers and copolymers with fluorescence properties were successfully synthesized; however, it was not possible to influence the fluorescence by the pH-depending dimerization of the zwitterion.

The third project of this work was focused on the application of GCP-polymers as non-viral vectors for gene transfection. Three new GCP-monomers were synthesized and afterwards converted into homopolymers or to copolymers in combination with M4. Two of the homopolymers showed first positive results in gene transfection experiments with low efficiency. Due to the combination with the monomer M4 the solubility in water as well as the efficiency of the GT1 based vector were distinctly improved. Besides the combination with other monomers, the modular approach allows to easily change the monomer ratios. Increase of the GT1 amount resulted in even higher transfection efficiencies. However, an increase of the M4 monomer results in lower efficiencies and additionally in higher cell toxicity.

  • The new developed GCP-Copolymers are suitable for the application as non-viral vectors in gene delivery, and it was possible to systematically optimize the vectors with the modular approach.
