MD-Simulation von PNIPAM in Wasser - Vergleich von Samplingmethode und Kraftfeld

Im Rahmen dieser Arbeit wurde Poly(N-isopropylacrylamid) (PNIPAM) als Pentamer und 30mer in Molekulardynamiksimulationen mit und ohne Umbrella Sampling oberhalb und unterhalb der oberen kritischen Löslichkeitstemperatur (LCST) untersucht. Zur Beschreibung des Wassers wurde das TIP3P-Kraftfeld verwendet, für PNIPAM beider Kettenlängen das AMBER-Kraftfeld und für das 30mer PNIPAM verschiedene Varianten des OPLS-Kraftfeldes. Untersucht wurden der Einfluss der Temperatur auf den Gyrationsradius als Kenngröße für Lösungseigenschaften und den Phasenübergang. Im AMBER-Kraftfeld werden zwei stabile Gyrationsradien beobachtet, von denen der kleinere oberhalb der LCST bevorzugt wird, der größere unterhalb. Mit höherer Temperatur werden zunehmend weniger Wasserstoffbrückenbindungen zum umgebenden Wasser ausgebildet, was zu einer Destabilisierung der größeren Gyrationsradien führt. Die Zahl der koordinierten Wassermoleküle ist konstant, die Hydrathülle ist bei höherer Temperatur in gleichem Maße vorhanden, allerdings weniger geordnet. Die Änderung anderer Eigenschaften wie die Formänderung von prolat zu sphärisch oder die stärkere Ausrichtung der Seitenketten weg vom Polymerzentrum ist über den Gyrationsradius nur mittelbar mit der Temperatur verknüpft. Die Simulationen mit dem OPLS-Kraftfeld zeigen, dass die Modellierung der Torsionspotentiale wichtig für die Ausbildung der temperaturabhängigen Wasserstoffbrückenbindungen wie auch für die Änderung des Gyrationsradius sind. Nur durch den Übergang von trans- zu gauche-Konformationen kann der Phasenübergang geschehen. Es wurden Simulationen mit und ohne Umbrella Sampling durchgeführt. Beim Umbrella Sampling wird statt einer Simulation ein Set von Simulationen durchgeführt, bei denen der Gyrationsradius jeweils harmonisch um einen Gleichgewichtswert eingeschränkt wird. Anschießend wird der Einfluss des Umbrellapotentials wieder kompensiert. Beide Arten der Simulation zeigen zwar ähnliche Verteilungen für den Gyrationsradius, allerdings schränkt das Umbrella Sampling die ohnehin schon langsame Konformationsänderung des Polymers weiter ein, sodass Simulationen ausgehend von kollabierten Formen die lokalen Minima nicht überwinden können. Schlussfolgerung ist also, dass Umbrella Sampling zur Betrachtung des PNIPAMs nur bedingt geeignet ist. Die Hydrathülle wird nicht durch das Umbrella Sampling beeinflusst.

A pentamer and a 30mer of poly(N-isopropylacrylamide) (PNIPAM) in aqueous solution was investigated using Molecular Dynamics Simulations and Umbrella Sampling above and below the lower critical solution temperature (LCST). The TIP3P force field was used for the description of water. Both PNIPAM molecules were simulated using the AMBER force field and the 30mer PNIPAM was investigated using different versions of the OPLS force field. The temperature effect has been investigated by means of the radius of gyration as a parameter for solvation properties. Two stable radii of gyration were found using the AMBER force field, the smaller one was favored above LCST, the larger one below. With increasing temperature less hydrogen bridges were formed. This destabilizes the larger radii of gyration. The number of coordinated water molecules is constant so the hydration shell is similar at all temperatures but less ordered at higher temperature. Other properties like the change in shape from prolate to spherical or the orientation of the sidechains away from the center of mass at higher temperature are mediated by the radius of gyration and thereby temperature sensitive. The simulations using the OPLS force field showed that the choice of the torsion potential is important for the formation of the temperature sensitive hydrogen bonds as well as for the change of the radius of gyration. The transition from trans to gauche conformations enables the phase transition. Simulations with and without Umbrella Sampling have been employed. When Umbrella Sampling was used, a set of simulations for the same system has been performed instead of one single simulation. Within the set of simulations the radius of gyrations was harmonically restrained at an equilibrium value ranging between 0.7 and 2.0 nm. The bias of the Umbrella Potential was compensated afterwards. Both simulation methods showed similar distributions for the radius of gyration, but the Umbrella Sampling restricts the already slow conformational change of the polymer. This leads to the fact, that simulations starting from a collapsed conformation of the polymer cannot overcome the local energy barrier. The hydration shell is not influenced by Umbrella Sampling. Thereby Umbrella Sampling is only partly suitable for the investigation of PNIPAM.
