Neuroblastome entstehen aus Neuralleistenstammzellen

Das Neuroblastom ist der häufigste solide extrakranielle Tumor des Kindesalters und weist eine große klinische Heterogenität auf. Amplifikationen des MYCN Onkogens finden sich in 20% aller Neuroblastome und korrelieren mit schlechter Prognose. Es wird diskutiert, dass sich Neuroblastome aus unreifen Neurralleistenstammzellen entwickeln. Um zu untersuchen, ob sich Neuroblastome tatsächlich aus Neuralleistenstammzellen entwickeln, wurde im Rahmen dieser Arbeit die Neuralleistenstammzellinie JoMa1 verwendet. JoMa1 Zellen wurden mit MYCN oder mit mutierten ALK-1174, einem Onkogen, welches in ca. 10% aller Neuroblastome mutiert ist, transfiziert. Beide Onkogene immortalisierten JoMa1 Zellen in vitro. JoMa1-MYCN und JoMa1-ALK-1174L Zellen, nicht aber JoMa1 Kontrollzellen, bildeten Tumore in Nacktmäusen, die humanen Neuroblastomen glichen. Die serielle Transplantation dieser Tumoren führte zu einer Beschleunigung der Tumorentwicklung. Eine Behandlung einer Zellinie, die aus einem JoMa1-MYCN Tumor etabliert worden war, mit einem MYCN Inhibitor bestätigte, daß die Tumorzellen weiterhin abhängig von MYCN waren. Im Rahmen dieser Arbeit konnte nachgewiesen werden, daß sich Neuroblastome tatsächlich aus Neuralleistenstammzellen entwickeln. Es konnte darüber hinaus gezeigt werden, daß die Onkogene MYCN und ALKF1174L tatsächlich das Potential haben, die Neuroblastomentstehung zu initiieren. Außerdem wurde mit dem JoMa1-Neuroblastommodell ein System zur Analyse der frühen Schritte der Neuroblastomentstehung und zur raschen Analyse des Neuroblastom-initierenden Potentials von Genen bzw. Genmutationen etabliert.

Neurablastoma are the most common solid extracanial tumors of childhoodness and show a great clinical heterogenecity. Amplifications of the oncogene MYCN occur in 20% of all neuroblastoma and are connected with a poor prognosis. It is postulated that neuroblastoma originate from neuralcreststemcells. To prove whether neuroblastoma really originate from neuralcreststemcells JoMa1 cells - murine neuralcreststemcells - were used. JoMa1 cells were transfected with MYCN or mutated ALK-F1174. ALK-F1174 is an onocogene which is mutated in about 10% of all neurablastoma. Both oncogenes immortalize JoMa1 cells in vitro. In contrast to untransfected JoMa1 controll cells, JoMa1-MYCN and JoMa1-ALK-F1174L cells formed tumors in nude mice. These tumors resembled human neuroblastoma. The serial transplantaion of these tumors led to an increase of tumor development. Treatment of one cell line that had been established from a JoMa1-MYCN-tumor with an inhibitor of MYCN endorsed the remaining addiction of these tumors to MYCN. Summing up it could be shown that neurablastoma originate from neuralcreststemcells. Also it could be shown that the oncogenes MYCN and ALK-F1174L have the potential to initiate the formation of neuroblastoma. In addition, with the JoMa1 model a system has been established to analize the early steps of genesis of neuroblastoma and to analyze the oncogenic and neurablastoma-initiating potential of - partly yet unknown - genes and gene-mutations in a fast way.
